The first a pastoral opera, L ' amore innocente ( Innocent Love ) was a light hearted comedy set in the Austrian mountains, and the second was based on an episode from Cervantes Don Quixote – Don Chisciotte alle nozze di Gamace ( Don Quixote at the Marriage of Camacho ).

Don Chisciotte was a mix of ballet and opera buffa, and the lead female roles in L ' amore innocente were designed to contrast and highlight the different traditions of operatic writing for soprano, even borrowing stylistic flourishes from opera-seria in the use of coloratura in what was a short pastoral comedy more in keeping with a Roman Intermezzo.

With the Neapolitan singer Mariano Apicella he wrote two Neapolitan song albums: Meglio ' na canzone in 2003 and L ' ultimo amore in 2006.

In 1986 Nespolo had the possibility of designing the stage sets for Ferruccio Busoni ’ s opera Turandot at Connecticut Grand Opera, Stamford, the first of several theatre works he made in the following years, such as the sets and costumes for Paisiello's Don Chisciotte at the Teatro dell ' Opera di Roma in 1990 and for Donizetti's L ' elisir d ' amore, a production for the Rome Opera, the Paris Opera and the Opéra de Lausanne, Liège and Metz in 1995.

His best-known works are the operas L ' elisir d ' amore ( 1832 ), Lucia di Lammermoor ( 1835 ), and Don Pasquale ( 1843 ), all in Italian, and the French operas La favorite and La fille du régiment ( both from 1840 ).

L ' elisir d ' amore, a comedy produced in 1832, came soon after, and is deemed one of the masterpieces of 19th-century opera buffa ( as is his Don Pasquale, written for Paris in 1843 ).

Shortly after L ' elisir d ' amore, Donizetti composed Lucia di Lammermoor, based on the Sir Walter Scott novel The Bride of Lammermoor.

It was recognized at the time as, and is still considered, Donizetti's comic masterpiece, and remains one of the most popular of his 66 operas, as well as one of the three most popular Italian comic operas, the other two being Rossini's The Barber of Seville and Donizetti's own L ' elisir d ' amore.
