Hamilton married in 1532, to the Lady Margaret Douglas, daughter of James Douglas, 3rd Earl of Morton and Catherine Stewart, herself a natural daughter of James IV.

In February 1532 he, Lord Oliphant, and various barons in that district of Scotland were fined for not appearing to sit as jurymen at the trial of Lady Glamis at Forfar for poisoning her husband.

Anne had become pregnant by the end of 1532 and gave birth on 7 September 1533 to Elizabeth named in honour of Henry's mother.

Anne and Henry attended a meeting with the French king at Calais in winter 1532, in which Henry hoped to enlist the support of Francis I of France for his intended marriage.

She became a maid-of-honour in 1532 to Queen Catherine, but Jane may have served Catherine as early as 1527, and went on to serve Queen Anne Boleyn.

In 1532, Henry VIII was preparing to repudiate Catherine of Aragon and marry Anne Boleyn, in defiance of the pope.

It may have taken place when Anne was with the King in Calais in November 1532, but it seems more likely that it took place at a secret ceremony on 25 January 1533.

In 1532 he accompanied his first cousin Anne Boleyn, the King, and the Duke of Richmond to France, staying there for more than a year as a member of the entourage of Francis I of France.

On 1 September 1532, a few months prior to her marriage to Henry VIII, Anne was granted the Marquesate of Pembroke ; she was found guilty of treason and executed in May 1536, at which point the title became either forfeit or extinct at her death without male children.

In 1532 Percy, now Earl of Northumberland stood in great danger, when his wife accused him of a precontract ( i. e. betrothal with legal force ) with Anne Boleyn.

On Sunday, 1 September 1532, Anne Boleyn was granted the Marquessate of Pembroke and land, mostly in Wales, worth over ₤ 1, 000.

Margaret was one of Anne's chief ladies-in-waiting, and accompanied her to Calais, France in 1532, where it is presumed Anne and Henry VIII made secret plans to marry in the immediate future.
