Ivan was the son of Vasili III and his second wife, Elena Glinskaya.

At first, his mother Elena Glinskaya acted as regent, but she died of what many believe to be assassination by poison when Ivan was only eight years old.

The boyars suggested that he take a new wife, and despite much opposition from the clergy, he divorced his barren wife and married Princess Elena Glinskaya, the daughter of a Serbian princess and niece of his friend Michael Glinski.

His niece Elena Glinskaya was married to Vasily III, Grand Prince of Moscow, and Ivan the Terrible was her son.

He was freed after his niece Elena Glinskaya married Vasili III in 1526.

Glinski spent 12 years in prison until 1526, when Vasili III married his niece Elena Glinskaya.

Upon the death of Vasily III's widow, Elena Glinskaya, he challenged the authority of Prince Ivan Belsky, procured his incarceration, married Anastasia of Kazan ( Ivan III's granddaughter ), and proclaimed himself regent for Vasily III's heir, the young Ivan IV, in 1538.

During the regency of Elena Glinskaya he served as the governor of Moscow and of Kholmogory.

It was later rumoured, that Elena brought witches from Finland and people of the Sami to help her conceive by the help of magic On his deathbed, Vasili III transferred his powers to Elena Glinskaya until his oldest son Ivan was mature enough to rule the country.

Elena Glinskaya challenged the claims of her brothers-in-law, Yury of Dmitrov and Andrey of Staritsa.

According to other accounts, he was involved in the conspiracy of Prince Andrey of Staritsa against Elena Glinskaya and, when their plans were discovered, he escaped to Solovetsky Monastery on the White Sea.

After 40 days of mourning, Andrey applied to his widow Elena Glinskaya for extension of his demesnes.
