During the reign of her cousin Anna ( 1730 – 1740 ), Elizabeth was gathering support in the background ; but after the death of Empress Anna, the regency of Anna Leopoldovna with infant Ivan VI was marked by high taxes and economic problems.

Elizabeth was only too aware that the deposed Ivan VI, whom she had imprisoned in the Schlusselburg Fortress and placed in solitary confinement, was a threat to her throne.

According to historian Robert Nisbet Bain, it was one of Elizabeth ’ s “ chief glories that, so far as she was able, she put a stop to that mischievous contention of rival ambitions at Court, which had disgraced the reigns of Peter II, Anne and Ivan VI, and enabled foreign powers to freely interfere in the domestic affairs of Russia .” She was also deeply religious, passing several pieces of legislation that undid much of the work her father had done to limit the power of the church.

As her health declined she declared her grandnephew, Ivan VI, should succeed her, and appointed Biron as regent.

As a consequence, shortly after Anna's death Elizabeth Petrovna, Peter I's legitimized daughter, managed to gain the favor of the populace, locked Ivan VI in a dungeon and exiled his mother.

By his wife Irene Asanina, a daughter of Andronikos Asan ( son of Emperor Ivan Asen III of Bulgaria by Eirene Palaiologina, herself daughter of Emperor Michael VIII Palaiologos ), John VI Kantakouzenos had several children, including:

Her son and Ivan's great-grandson, Ivan VI would be the last Russian emperor among the issue of Maria Miloslavskaya, the first wife of Tsar Alexis.

In fact, with Ivan VI ( who was murdered and buried in the fortress of Shlisselburg ), he is the only post-Petrine monarch not buried in the Peter and Paul Cathedral in Saint Petersburg.
